Automated Bioanalytical SPE

This basic method provides the core methodology for translating a workflow into OneLab-executable script(s) as an attempt to fully or semi-automate a specific procedure. It demonstrates the benefits of automation and highlights OneLab capabilities and best practices to promote solution adoption, helping transition from manual to a more automated approach. It can be used alone or serves as a building block for a more complex workflow and is easily adaptable to users' requirements.
Overview
Fully Automated Bioanalytical SPE Sample Preparation, using the Extraction+ Connected Device with the Andrew+ Pipetting Robot
Sample preparation, which isolates target analytes from the matrix and removes as many interferences as possible, is a critical component of the bioanalytical workflow (Figure 1). Introducing automation to the bioanalytical lab helps to minimize or eliminate some of these challenges. In this application, the Andrew+ Pipetting Robot was used to generate calibration curves and QC samples using a diversity of small molecule pharmaceutical drugs in plasma. The Andrew+ Pipetting Robot (Andrew+) and the new Extraction+ Connected Device were then used to perform solid phase extraction (SPE) of these prepared samples in both 1 cc cartridge and 96-well macro plate formats.

The photo below displays the Andrew+ Pipetting Robot configured with the Extraction+ system along with labware Dominos, Andrew Alliance Bluetooth electronic pipettes and a plate gripper on the tool stand. the Extraction+ (#5) includes the connected vacuum pump, flow-through waste container, the vacuum manifold with the collar, and the integrated collar lifter. Here in this photo, as an example, 6 cc SPE cartridges are loaded into a specific 12-position adaptor and placed onto the Extraction+ manifold.

Protocols and their corresponding visual Andrew+ deck layouts used for the bioanalytical plate and cartridge SPE sample preparation are described below. For the SPE plate format, a single protocol was used for sample preparation followed by two separate tutorial protocols for SPE extraction performed with Andrew+ configured with Extraction+ in which samples were either processed row or column-wise. For the SPE 1 cc cartridge format, a single protocol was used for sample preparation followed by one separate protocol for SPE extraction performed with Andrew+ configured with Extraction+.
